Statins and Ibrutinib in CLL/SLL: Emerging Evidence for Combination Therapy

The observation that CLL cells are unusually dependent on cholesterol metabolism has generated sustained interest in whether HMG-CoA reductase inhibitors — statins — might enhance the activity of BTK inhibitors such as ibrutinib. This article reviews the mechanistic rationale, summarises the available preclinical and early clinical data, and explains the design of our KCLG-HAEM-01 Phase II trial.

For clinicians managing patients with relapsed/refractory CLL on ibrutinib, the potential for a well-tolerated, widely available adjunct therapy makes this a clinically important question — one we are now formally testing.
